چکیده

Factors affecting inhibition of myosin ATPase by ATP, such as concentration of the enzyme, of ATP, and of the divalent metals, calcium and magnesium, have been studied and the results are reported here. Experiments were carried out under conditions in which no known magnesium effects occurred, so that only the effect of calcium on the nucleoside triphosphatase activity of myosin A could be qualitatively characterized. Myosin A ATPase was inhibited by free ATP at 0.15 ionic strength in the presence of millimolar calcium and 0.006 M KCl. A reaction model is proposed in which Ca-ATP is the substrate for native myosin A and inhibition is brought about by 2 moles of ATP.

The Effect of Vitrification and in vitro Culture on the Adenosine Triphosphate Content and Mitochondrial Distribution of Mouse Pre-Implantation Embryos

Background: The mitochondria are an important source of adenosine triphosphate (ATP) production in pre-implantation embryo. Therefore, the objective of this study was to investigate the effect of vitrification and in vitro culture of mouse embryos on their mitochondrial distribution and ATP content. Methods: The embryos at 2-PN, 4-cell and blastocyst stages were collected from the oviduct of st...

An Adenosine Triphosphate Bioluminescence Method for Evaluating the Microbial Contamination of the Salad-Preparing Tables and Salad-Serving Dishes in Restaurants of Mashhad City, Iran

Introduction: Consumption of vegetable products is increasing commonly in the world because they are recognized as an important source of nutrients, vitamins, and fiber for humans. Salads are among the most widely used foods that are also known as the most contaminated foods in restaurants. This study was conducted to determine the microbial contamination of salad-preparing tables and salad-ser...

Comparing Human Sperm Quality Preserved at Two Different Temperatures; Effect of Trolox, Coenzyme Q10 and Extracellular Adenosine Triphosphate

Cooling method was proposed to maintain the sperm quality for several days. Nevertheless, during this procedure, sperm is encountered to “cold shock”, and its quality decreases time-dependently. This study was designed to improve the in vitro sperm preservation methods. Thirty normal semen samples were examined in Shiraz, Iran, 2017. Fifteen samples were incubated at 22-27 °C and 15 samples wer...

Kinetics and substrate binding of myosin adenosine triphosphatase.

The rate of hydrolysis of adenosine triphosphate and the binding of inorganic pyrophosphate to myosin vary inversely with increasing concentration of free Mg++, in the presence of a magnesium ethylenediaminetetraacetate buffer system; half-maximum binding of pyrophosphate and 50% inhibition of adenosine triphosphatase activity occur at a free Mg++ concentration about 10M7 M at 3O. The binding o...

The active site of myosin adenosine triphosphatase. I. Localization of one of the sulfhydryl groups.

It is now widely recognized that under certain conditions of temperature and pH (1, 2)) the Ca++-activated hydrolysis of adenosine triphosphate (ATP) by myosin exhibits a biphasic response to titration of the sulfhydryl groups of the protein by organic mercurials and by N-ethylmaleimide (3-5). On the other hand, titration by these reagents results only in inhibition if inosine triphosphate or g...
